COMPARISON OF TWO METHODS OF DNA EXTRACTION IN PARASITES (Sarcoptes scabiei) OF HUACAYA ALPACAS FROM THE PROVINCE OF HUANCAVELICA

Abstract

The objective of this work was to compare two DNA extraction methods (Phenol – Chloroform and Quick-DNA™ 96 Plus Kit) in parasites (Sarcoptes scabiei) of Huacaya alpacas in terms of quantity and quality (purity and integrity). For this, 18 alpacas were selected, from which samples of the skin lesions caused by scabies were extracted. Subsequently, the isolation of the parasites and the extraction of DNA were carried out with the two extraction methods mentioned. For data analysis, descriptive statistics and the Mann-Whitney U Test were used, using the R statistical program. Obtaining the following results: the amount of DNA for the Phenol – Chloroform (F-C) method was 6.4 ± 1.2 ng/ ul and for the Quick-DNA™ 96 Plus Kit (Quick) method it was 6.4 ± 1.6 ng/ul, not finding significant statistical differences (P>0.05), regarding the range of the DNA purity index (A260/A280 ), was between 1.8 ± 0.1 nm and 1.9 ± 0.1 nm for the F-C method and the Quick method respectively, not finding significant statistical differences (P>0.05); Regarding completeness, regular completenesses were observed in both methods. The amount of DNA obtained with the two extraction methods is small, but useful for molecular studies. As for quality, it is within the optimal range. Both extraction methods are similar with respect to the quantity and quality of the DNA.
